Today there is no standardized protocols for freezing, thawing and umbilical cord blood (UCB) stem cells preparation for transplantation. We studied 31 units of cord blood- 11 thawed units were washed using Sepax S-100 (Biosafe, Switzerland) (group 1), 11 units were washed using centrifuge (group 2) and 9 unpwashed units were reference group (group 3).
The study includes enumeration of nuclear cells, amount and viability of CD34/CD45+ cells in UCB units before cryopreservation, after thawing and washing with two methods mentioned above. Median of CD34/45+ cells recovery in groups 1, 2 and 3 was 92±9%, 81±7% и 94±6%, respectively, median of nuclear cells recovery in groups 1, 2 and 3 was 93±3%, 85±4% и 94±3%, respectively and viability in groups 1, 2 and 3 was 94,9±2,4%, 93,7±2,2% и 94,4±2,1%, respectively. Our conclusion is that method of thawing and subsequent processing of UCB unit doesn’t affect viability of CD34/45+ cells and the largest nuclear and CD34/45+ cells lost (15% and 19%, respectively) observed in group 2, while parameters of cell concentrate didn’t differ significantly. Therefore in case of availability of appropriate technical equipment, washing UCB unit from cryoprotectant using an automated system does not lead to deterioration of quality parameters of UCB unit.
