More than 100,000 joint replacement surgeries are performed annually in Russia, but the medical steel of endoprostheses currently used can have a toxic effect on humans, so we conducted in vitro testing on mesenchymal stromal cells from human bone marrow for biocompatibility and cytotoxicity of medical steel prototypes. The object of the study were prototypes of an alloy of medical steel obtained by machining on a milling machine. In the process of work, a comprehensive assessment of the biocompatibility and cytotoxicity of the provided prototypes on mesenchymal stromal cells from human bone marrow was carried out. The following methods were used in the study: culture, fluorescence microscopy using the LIVE/DEAD Viability/Cytotoxicity Kit fluorophores (Life technologies USA); biochemical (MTT-test). The culture of mesenchymal stromal cells was obtained from human bone marrow, after signing an informed voluntary consent. The results of the testing of experimental samples indicate the absence of cytotoxicity. A greater number of viable cells and evenly distributed over the surface was observed on a smooth surface. The number of viable cells in the MTT test was 90.5%. The investigated alloy of medical steel is not cytotoxic. The adhesion of cells directly depends on the nature of the surface of the material, greater adhesive activity is noted on smooth areas than on rough ones. The obtained results of in vitro studies can be used to improve the technology for the development of biocompatible implants with good opportunities for osseointegration
