The effect of violuric acid on the antioxidant system in the long-term period after poisoning with sodium thiopental

Brief summary

The antioxidant system, in particular the glutathione system, plays a leading role in the protection of the cell from the damaging effects of reactive oxygen species and in maintaining the homeostatic balance in the cell. Structural and functional disorders at the cellular and subcellular levels, which underlie the pathogenesis of many diseases, are a direct consequence of the accumulation of free radicals and the subsequent decrease in the antioxidant status. Thus, a comprehensive assessment of the state of the antioxidant system and the activity of its enzymes can provide a primary idea of free radical homeostasis against the background of one or another exposure. It is important to study changes in its activity in the long-term consequences of acute poisoning with neurotoxicants, as well as to identify promising drugs for the pharmacological correction of the revealed pathological changes. This article investigated the effect of a derivative of violuric acid on the antioxidant system in rat erythrocytes 1 and 3 months after sodium thiopental poisoning. As a result of the experimental study, a shift in the balance between the pro- and antioxidant systems was revealed in the long-term period after intoxication in favor of the former. The reasons for the violation of this balance are associated not only with the activation of free radical processes, but also with the depletion of the antioxidant system, namely, a decrease in the level of reduced glutathione and a decrease in the activity of antioxidant enzymes. The use of the zinc complex of 1-butylvoluric acid made it possible to eliminate the revealed imbalance due to the activation of enzymes of the antioxidant system and a decrease in the activity of free radical oxidation, which indicates the possibility of its use for the treatment of long-term effects of acute poisoning with neurotoxicants.
