Colonyforming capcity of haematopoietic stem cells and cd34+ cells number in the product of apheresis after different mobilization regimens in patients with multiple myeloma

Brief summary

The aim of the work was to compare colonyforming capacity and CD34+cells number in the product of aphaeresis received in patients with multiple myeloma (MM) after mobilization regimens (MR) with cyclophosphamide (CY) or vinorelbine. Methods. Data of 71 MM patients were retrospectively analyzed. The patients age was 26-67 years. Two groups of patients were formed according to the MR variant: with CY (group 1) and with vinorelbine (group 2). Aphaeresis was initiated when CD34+ cells number was ≥20/1 μL of peripheral blood. MethoCult H4435 medium was used to study colonyforming capacity. CD34+ cells were determined by using flow laser cytofluorimeter "Cytomics FC 500" ("Beckman Coulter", USA). Conditioning regimens used before autotransplant infusions were next: Mel200, Mel140, BEAM, or combination of melphalan with thiothepa. Statistical processing of the data was carried out using standard statistical methods. Results. According to the cultural research there was no any difference in the colonyforming capacity of aphaeresis product getting after MR with CY or vinorelbine. The intensity of MP did not influence on the CD34+ cells number too: 1.67х109/L and 1.68х109/L accordingly; р>0.05. Conditioning regimen Mel200 was used more often for the first autotransplantation and regimen Mel140 was used more often for the second one. In the cases with CY used for RM autografting of neutrophils was faster after the second transplantation than after the first one: 9 and 10 days accordingly; р=0.0007. If vinorelbine was used for RM there was no difference in durability of posttransplantation agranulocytosis after the first and the second transplantation. Conclusion. The capacity of CY and vinorelbine used for stem cell mobilization from bone marrow to peripheral blood are equivalent.
