Assessement of genome stability at the personnel of plant "Khimzavod" and the population in an area of its location

Brief summary

Abstract Professional activity of the personnel of "Khimzavod" plant, which performs disposal of liquid-fuel rocket engines, is a danger of contamination by rocket fuel components (RFC) of the objects of production environment of "Khimzavod" plant and the surrounding territory. The most toxic pollutants are the main RFC - 1.1-dimethylhydrazine (heptyl, unsymmetrical dimethylhydrazine - UDMH), and nitrosodimethylamine (NDMA), a product of decomposition of UDMH under the influence of oxygen and moisture in the surrounding air. Aim of the study. The purpose of this study was to assess the genome stability of "Khimzavod" personnel and the population in an area of its Materials and methods. The subjects of this study were human white blood cells. Two biomarkers were used to assess genome instability: - Primary DNA damage (single breaks, double breaks, and AP sites) determined by Comet assay. - Mutations (chromosomal aberrations and aneuploidy) determined in the culture of lymphocytes with cytokinetic block. The third biomarker (index of proliferation - IP) was allowed to register the cytotoxic action of RFC. Two groups of "Khimzavod" personnel were formed: those who had direct contact with RFC and did not work with RFC, as well as two groups of people living in Podgorny and Zheleznogorsk. Results. It was found that the degree of primary damage to the nuclear DNA of leukocytes and the frequency of occurrence of micronuclei in dual nucleus lymphocytes in all groups studied did not differ from each other. This indicates the absence of genome stability disorders in the personnel of the "Khimzavod" plant and in the population living in Zheleznogorsk and Podgorny settlement. The index of proliferation in the lymphocyte culture in the population of Podgorny settlement is significantly higher than in the population of Zheleznogorsk, which indicates a more favorable environmental situation in the area of the plant "Khimzavod". When comparing the values of IP lymphocytes of the population of Podgorny settlement with the personnel of the plant "Khimzavod", a statistically significant decrease in the proliferative activity of lymphocytes in the group of personnel in contact with RFC was obtained. The value of PI of lymphocytes in the personnel who do not work with RFC, occupied an intermediate value. Besides, in the group of personnel of "Khimzavod" enterprise who had contact with RFC, a high degree of correlation between the value of IP of lymphocytes and length of service was found. The Pearson correlation coefficient was -0.567 (p = 0.027). Based on the obtained data of IP lymphocytes in the Podgorny settlement group, the reference interval of the norm was calculated. Its value for this region was 1.60 - 1.88. When comparing with the reference interval of the norm of IP of lymphocytes calculated for the group of population of Podgorny settlement, two cases of reduction of IP of lymphocytes below the norm were revealed among the personnel of the plant "Khimzavod" contacting with RFC (1.59 and 1.51). Conclusions. Thus, on the basis of the obtained results it is possible to draw a conclusion about the absence of genome stability disorders at the personnel of plant "Khimzavod" and the population in an area of its location. In the group of personnel of "Khimzavod" enterprise associated with the disposal of liquid-fuel rockets, a decrease in the proliferation index of lymphocytes was found, which indicates the toxic effect of 1,1-dimethylhydrazine (heptyl).
