Early changes in urinary bladder barrier function in cyclophosphamide myeloablative chemotherapy rat model

Brief summary

The subject of the study was the hypothesis of the compromised urinary bladder barrier function in cyclophosphamide (CY) myeloablative chemotherapy. In intact rats, the intravesical instillation of ammonium acetate or urea solutions (10-fold concentrated compared with higher ammonia or urea urine levels) or 1% Evans blue dye (EB) revealed the permeability of urothelium to ammonia or EB, but not to urea. CY promoted the flux of EB into the blood and, to a lesser extent, the accumulation of EB in brain and visceral organs, increased the permeability of urothelium to urea, but not to ammonia. Intravesical administration of urea promoted the CY-induced hyperammonaemia. Involvement of ammonia produced by the increased urea recycling, determines the clinical relevance of these data.
